The Hallmarks of Aging (López-Otín et al., 2013; updated 2023 to 12 hallmarks) provide the conceptual framework: genomic instability, telomere attrition, epigenetic alterations, loss of proteostasis, disabled macroautophagy, deregulated nutrient sensing, mitochondrial dysfunction, cellular senescence, stem cell exhaustion, altered intercellular communication, chronic inflammation (inflammaging), and dysbiosis. Each hallmark has been intervened upon in animal models with life extension results.

- **Yamanaka factors (2006)**: Oct4, Sox2, Klf4, c-Myc reprogram somatic cells to pluripotency. Partial reprogramming (cycled, without full dedifferentiation) reverses aging phenotypes in mice (Belmonte lab, Salk Institute) and restores vision in old mice (Sinclair lab, Harvard).
- **Senolytics**: drugs (dasatinib + quercetin, navitoclax) that selectively kill senescent cells extend healthspan and lifespan in mice; human trials ongoing (Unity Biotechnology, Oisín Biotechnologies).
- **Epigenetic reprogramming**: the Horvath clock gives a quantitative biological age; interventions that reverse it in blood are being tested.
